Abstract: Elvis Presley with his song “Seeing Is Believing” was among the many who picked
the idea of visual perception role in the nature observation. Today, with the
development of advanced genetically encoded reporters – the imaging of
miscellaneous molecular/cellular events becomes key driver in answering the
biological questions. Electron microscopy (EM) together with super-resolution
microscopy (SRM) develop into the methods of choice due to the unprecedented
resolution power, needed for molecular cell biology. Being limited in reporters for
SRM, researches face even more severe lack of the similar tools for EM. Studying
the vast combinatory space of protein structures – it is feasible to circumvent this
limitation through the means of purposeful protein design and special sample
prep/imaging modalities. In view of this, the goal of the current work is to develop
and characterize the genetically controlled reporters, suitable for filling the toolbox
gap in EM and simultaneously applicable in SRM.
