Abstract: Fluorescent reporters like GFP kicked off a new era in biomedical research by enabling easy fusion to a variety of proteins and enzyme targets for multi-color readouts of cellular processes. Still, for imaging at the best resolution achievable today through Electron Microscopy (EM), equally flexible reporters are in comparison yet lacking. Advances in protein structure predictions and design in the recent years have been fuelled by technological breakthroughs, extensive growth of biological databases and increasing availability of computational power. Using such tools we aim to introduce a set of genetically encoded nanostructures that are able to generate geometrically distinct and precise EM contrast.
