Abstract: Recent breakthroughs in tissue-clearing methodologies have transformed the study of intact tissues, organs, and organisms, offering a 3D visualization of interconnected systems at a single-cell resolution. These methods, integrated with light-sheet microscopy, AI-based analysis, and proteomics, streamline histological and molecular examinations, potentially reducing processing time and cost. The chemical processes enable whole-organ labeling, broadening the scope to include thick human tissues and facilitating comprehensive whole-body scanning of small mammalian models. By harnessing this synergy, researchers can rapidly extract comprehensive, multiscale insights from complex biological specimens. This seminar will provide an overview of cutting-edge tissue-clearing approaches, emphasizing the versatile DISCO protocols. Common implementation challenges and solutions will be addressed to equip attendees with the necessary knowledge to adopt these technologies seamlessly. Participants will gain valuable insights into using these technologies for unbiased, systems-level investigations spanning entire organs and organisms for an unprecedented understanding of biological processes to support disease research.
